研究目的：

通过多中心大样本随机对照试验，评价针刺改善PCOS排卵障碍的有效性和安全性，为针刺改善排卵障碍提供高质量临床证据。

Objectives of Study：

Through multicenter large-sample randomized controlled trials to evaluate the efficacy and safety of acupuncture in improving ovulation disorders in PCOS and to provide high-quality clinical evidence for the improvement of ovulation disorders by acupuncture.

药物成份或治疗方案详述：

Description for medicine or protocol of treatment in detail：

纳入标准：

（1）年龄21-40岁； （2）符合多囊卵巢综合征的诊断；稀发排卵或无排卵（OA）； （3）近一年内可自主行经，月经周期＞35天且≤60天； （4）患者自愿参加临床试验，并签署知情同意书。

Inclusion criteria

(1) Age 21-40 years; (2) Fits the diagnosis of polycystic ovary syndrome; sparse ovulation or anovulation (OA); (3) Can menstruate on their own within the past year with a menstrual cycle of >35 days and ≤60 days; (4) Patients volunteered to participate in the clinical trial and signed an informed consent form.

排除标准：

（1）库欣综合征、非经典型先天性肾上腺皮质增生、卵巢或肾上腺分泌雄激素的肿瘤、功能性下丘脑性闭经、甲状腺疾病、高泌乳素血症、早发性卵巢功能不全等可能引起高雄激素和排卵异常的疾病。 （2）先天性生殖器官发育异常、未处理的子宫内膜息肉、未处理的宫腔黏连、未治疗的宫腔感染、未处理的输卵管积水、薄型子宫内膜、子宫肌瘤（＞5cm、黏膜下子宫肌瘤）、子宫内膜异位症或其它器质性疾病； （3）近1个月内使用雌孕激素类药物及调脂降糖药； （4）近1月接受针灸、中药或中成药以调经助孕者； （5）合并严重心脑血管、肝、肾、恶性肿瘤、造血系统及精神疾病者； （6）BMI≥28kg/m²； （7）正处于妊娠或哺乳期者。

Exclusion criteria：

(1) Cushing's syndrome atypical congenital adrenocortical hyperplasia androgen-secreting tumours of the ovaries or adrenal glands functional hypothalamic amenorrhoea thyroid disorders hyperprolactinaemia early-onset ovarian insufficiency and other disorders that may cause hyperandrogenism and ovulatory abnormalities. (2) Congenital abnormalities of reproductive organ development untreated endometrial polyps untreated uterine adhesions untreated uterine infections untreated tubal hydrosalpinx thin endometrium uterine fibroids (>5cm submucosal fibroids) endometriosis or other organic diseases; (3) Use of estrogen-progestogen drugs and lipid-regulating and hypoglycemic drugs in the last 1 month; (4) Accepting acupuncture Chinese medicine or proprietary Chinese medicine to regulate menstruation and help pregnancy in the last 1 month; (5) Combined serious cardiovascular cerebrovascular hepatic renal malignant tumour haematopoietic system and psychiatric diseases; (6) BMI ≥ 28kg/m²; (7) Those who are in the period of pregnancy or breastfeeding. 随机方法（请说明由何人用什么方法产生随机序列）：

采用中央随机系统按1:1的比例将合格受试者随机分配至针刺组和假针刺组。随机化方案由统计软件SAS 9.4的PROC PLAN程序产生。中央随机系统由中国中医科学院中医临床基础医学研究所提供，随机分配方案及随机化过程中设定的各参数作为盲底由课题承担单位保存，并设置了严格的查看权限。通过电话或者网络将合格受试者的性别和出生日期输入中央随机系统，以获得每个患者的随机号和组别。此项研究为竞争性招募研究，患者将会竞争入组。竞争入组指的是所有研究中心互相竞争入组患者，直至达到总的目标值（患者的总人数）。

Randomization Procedure (please state who generates the random number sequence and by what method)：

A central randomisation system was used to randomly allocate eligible subjects to the needling and sham needling groups in a 1:1 ratio. The randomisation scheme was generated by the PROC PLAN program of the statistical software SAS 9.4. The central randomisation system was provided by the Institute of Clinical Basic Medicine of Traditional Chinese Medicine China Academy of Traditional Chinese Medicine and the random allocation scheme and each parameter set during the randomisation process were kept as a blind base by the subject undertaking unit with strict viewing privileges. The gender and date of birth of eligible subjects were entered into the central randomisation system by telephone or via the Internet to obtain the randomisation number and group for each patient. This is a competitive recruitment study and patients will compete for enrolment. Competitive enrolment means that all study centres compete with each other to enrol patients until the overall target (total number of patients) is reached.

盲法：

因针刺和假针刺干预方式不同，无法对针灸医师施盲。仅对受试者、疗效评价者及统计分析人员施盲。

Blinding：

It was not possible to blind the acupuncturists because of the difference between the needling and sham needling interventions. Only subjects efficacy evaluators and statistical analysts were blinded.
